An Energy Rich Phosphate from 2 - Methylimidazole through Nonenzymatic Photophosphorylation

چکیده

Sensitized photooxidation of imidazoles in aqueous solution and in the presence of inorganic phosphate afforded organic phosphates with yields up to 30%. The major phosphorylated product of oxidation of 2-methylimidazole was isolated and identified as the monophosphate of 4,5-dihydroxy-2-methyl-imidazoline with pKa values of 2.0, 5.2, and 9.8. The compound is energy rich in aqueous solution and hydrolyses in alkaline or neutral solutions irreversibly (A Hhyd = —9.8 kcal/mole). For the hydrolysis at pH 7 an activation-enthalpy of 25 kcal/mole and -entropy of 8 cal/deg.mole was observed. Be2+ ions catalyse the hydrolysis.

A Highly Reactive Imidazolium-Bridged Dinucleotide Intermediate in Nonenzymatic RNA Primer Extension.

Because of its importance for the origin of life, the nonenzymatic copying of RNA templates has been the subject of intense study for several decades. Previous characterizations of template-directed primer extension using 5'-phosphoryl-2-methylimidazole-activated nucleotides (2-MeImpNs) as substrates have assumed a classical in-line nucleophilic substitution mechanism, in which the 3'-hydroxyl ...

Nonenzymatie Formation of Energy Rich Phosphates in the Course of Imidazole Oxidation with Fenton's Reagent*

Oxidation of substituted imidazoles with modified Fenton's reagent (Fe(II), ascorbic acid, EDTA and H2O2) in aqueous solution at pH 5 -7 in the presence of inorganic orthophosphate gives high yields of an unknown bound phosphate, in the case of 2-methylimidazole up to 3 5 % based on inorganic phosphate. The dependence of the yields on reaction parameters and the hydrolytic behaviour of the comp...

Replacing Uridine with 2-Thiouridine Enhances the Rate and Fidelity of Nonenzymatic RNA Primer Extension

The nonenzymatic replication of RNA oligonucleotides is thought to have played a key role in the origin of life prior to the evolution of ribozyme-catalyzed RNA replication. Although the copying of oligo-C templates by 2-methylimidazole-activated G monomers can be quite efficient, the copying of mixed sequence templates, especially those containing A and U, is particularly slow and error-prone....

Energy Transformation in

Understanding metabolic energy transformation began with therealization of an 'intrusion' of phosphate into the mechanism of alcohoiicfermentation. The discoveryof an analogous participation of phosphate inmuscle glycolysis connected the metabolic generation of energy-rich phosphatebonds fed into a common transmitter, adenosine triphosphate (ATP), with theproduction of m...

Cyclic Photophosphorylation in the Mykotrophic Orhid Neottia nidus-avis.

The mykotrophic orchid Neottia nidus-avis (L.) Rich. is not able to evolve oxygen in the light. Plastid preparations from the lip (labellum) of the orchid perform a photosystem I-dependent photoreduction of methylviologen with the artificial electron donor couple 2,6-dichlorophenol indophenol ascorbate. Photosystem II reactions such as the ferricyanide Hill reaction or the photoreduction of 2,6...
